Recent events have brought far greater understanding of the threats that we all face from infectious agents. The COVID-19 pandemic has brought great suffering to the world, and even after recovery from an acute infection too many people experience prolonged harm. To further bolster research efforts to find treatments for long COVID, NIH launched a new component of the Researching COVID to Enhance Recovery (RECOVER) initiative(link is external) called RECOVER-Treating Long COVID (RECOVER-TLC). We convened a workshop to hear from more than 1,400 patients, advocates, health care providers and scientists to help inform this next phase of long COVID clinical trials. These trials will also be informed by extensive data from 40 pathobiology studies, review of 60 million electronic health records, and eight existing phase 2 clinical trials testing the safety and efficacy of 13 treatments that include drugs, biologics, medical devices and other therapies.

But the SARS-CoV-2 virus is continuing to wreak havoc on some Americans’ finances, according to a new study from the University of Georgia. The researchers found that long COVID-19 is making it harder for people to pay their bills, buy groceries and keep their utilities on. The study suggests much of that financial hardship is the result of lost jobs and reduced working hours. And the researchers found that the negative economic effects of the illness are present regardless of socioeconomic status. “COVID is still going on,” said Ishtiaque Fazlul, lead author of the study and an assistant professor in both UGA’s School of Public and International Affairs and UGA’s College of Public Health. “Long COVID is very much a problem that is affecting people’s lives right now. And it’s affecting people from all walks of life in terms of financial hardship.”

The Children and young people with Long Covid (CLoCK) study, published in Nature Communications Medicine and funded by the National Institute for Health and Care Research (NIHR), is the world’s largest longitudinal cohort study on long Covid in children. The researchers, led by Professor Sir Terence Stephenson and Professor Roz Shafran (both UCL Great Ormond Street Institute of Child Health), asked young people aged 11 to 17 about their health three, six, 12 and 24 months after taking a PCR test for the Covid virus between September 2020 and March 2021. They also asked them to recall their symptoms at the time of taking the test.

Long COVID: SARS-CoV-2 spike protein accumulation linked to long-lasting brain effects Peer-Reviewed Publication Helmholtz Munich (Helmholtz Zentrum München Deutsches Forschungszentrum für Gesundheit und Umwelt (GmbH)) Researchers from Helmholtz Munich and Ludwig-Maximilians-Universität (LMU) have identified a mechanism that may explain the neurological symptoms of Long COVID. The study shows that the SARS-CoV-2 spike protein remains in the brain’s protective layers, the meninges, and the skull’s bone marrow for up to four years after infection. This persistent presence of the spike protein could trigger chronic inflammation in affected individuals and increase the risk of neurodegenerative diseases. The team, led by Prof. Ali Ertürk, Director at the Institute for Intelligent Biotechnologies at Helmholtz Munich, also found that mRNA COVID-19 vaccines significantly reduce the accumulation of the spike protein in the brain. However, the persistence of spike protein after infection in the skull and meninges offers a target for new therapeutic strategies.

Impact Using online questionnaires validated during in-person medical examinations and including two different control groups, our study compellingly supports and adds to previous clinical observations in the field. Our study provides valuable insights into the prevalence and characteristics of pediatric LCS, highlighting the significant quality of life and functioning impairment compared to control groups. By detecting neutrophilic granulocyte dysfunction in children with LCS, we shed light on a previously overlooked pathophysiological component of the condition. We demonstrate a significant correlation between clinical symptoms and superoxide production, further enhancing our understanding of the underlying mechanisms of pediatric LCS.

CHICAGO – In patients with long COVID, lower pulmonary gas exchange may be associated with impaired cognitive function, according to a study being presented next week at the annual meeting of the Radiological Society of North America (RSNA). According to the National Center for Health Statistics, approximately 17.6% of adults in the U.S. have experienced a post-COVID condition commonly referred to as long COVID. People with long COVID may exhibit a wide variety of symptoms, including difficulty concentrating (“brain fog”), change in sense of smell or taste, fatigue, joint or muscle pain, dyspnea (shortness of breath), digestive symptoms, and more. These symptoms may persist for weeks, months, or even years after COVID-19 infection. Researchers from the University of Iowa in Iowa City set out to assess associations between pulmonary MRI gas exchange, structural and functional brain MRI, and cognition in long COVID patients. In pulmonary gas exchange, oxygen moves from the lungs to the bloodstream, while carbon dioxide moves from the bloodstream to the lungs. “This is the first time that MRI has been used to jointly assess lung and brain function to investigate their relationship in long COVID,” said the study’s lead author Keegan Staab, B.S., graduate research assistant in the Department of Radiology at the University of Iowa in Iowa City. “This research is new in that it combines multiple unique imaging types to study a multiorgan relationship in a disease population.”

Interpretation Younger and middle-age individuals are disproportionally affected by Neuro-PASC regardless of acute COVID-19 severity. Although older people more frequently have abnormal neurologic findings and comorbidities, younger and middle-age patients suffer from a higher burden of Neuro-PASC symptoms and cognitive dysfunction contributing to decreased QoL.
